If a bacterium cannot ferment glucose, what result will occur in a fermentation tube?
I assume you're talking about the Simmons' citrate test for your first question. [ If the bacterium can use the citrate on the media, then the pH will change, resulting in a change in media color from the nice green-ish color that the uninoculated media is to a dark blue. If the bacterum cannot use citrate as its primary C source, it will simply stay in lag phase, skip log phase, and basically
die. No food, death (Unless it forms endospores, in which case it might have time to throw a few of those ejection pods out before it dies off).
For Mannitol fermentation, I would use the MSA (Mannitol Salt) agar and streak it, the MCK agar (can't remember the acronym atm, sorry, I think it's like MacKonkey?) for lactose/sorbitol fermentation, BBL Phenol Red for adonitol, and TA agar for arabinose testing. ]
